[A] Budget: Looking for Proposals
[B] Scope of Service:
Vendor needs to provide cutting-edge transmission electron microscope system for high-throughput screening and high-resolution (better than 4 angstrom) single particle cryo-EM studies, as well as cryo-electron tomography of thin reconstituted preparations, cells, and tissues to the government authority located in Bethesda, MD.
- The microscope shall be capable of 200 KV accelerating voltage and equipped with a field emission
gun.
- The microscope must feature constant-power objective lenses to ensure maximum stability during long periods of data-collection.
- The microscope must feature a system for loading at least 12 cryo-specimens simultaneously to maximize the chance of successful data collection during each experiment and enable high-throughput screening of multiple sample conditions.
- The microscope must feature an internal stage system to minimize mechanical drift during data collection, as well as enabling high-tilt cryo-tomography.
- The microscope must be equipped with a direct-electron detector.
- The microscope must also be equipped with a high-speed phopho coated CMOS camera for live observation of samples with high-intensity beams as well as instrument alignment.
- The microscope must feature an integrated software package for automated single-particle and tomography data-collection capable of seamlessly controlling both the instrument and detector.
- The microscope must have an auto-loader compatible with the Krios microscope necessary for transferring the same Cassette between each microscope. Each cassette should accommodate up to 12 tem samples held in krios autogrids.
- The microscope must have a falcon III camera with EPU software.
- The microscope must have a phase plate with continuous film, controlled by epu and tomo data acquisition software.
